[Proteintech] 실험실에서 10분이면 충분한 Self Antibody Labeling Kits

FlexAble을 이용한 항체 labeling은 high affinity, non-covalent FlexLinker를 사용하여 만들어 집니다. FlexLinker는 몇 분 내에 항체에 단단히 결합하고, 한 번 결합하면 항체로부터 분리되지 않습니다. Unbound FlexLinker는 FlexQuencher를 추가하여 무력화 시킵니다.

Labeling reaction 중에는 응집이나 항체 손실이 관찰되지 않습니다. 원래의 항체 기능을 유지한 항체를 생성합니다.

FlexLinker 특징
FlexLinker는 Proteintech에서 개발한 물질로 형광 dye와 covalent 결합을 이루고 있습니다. 항체의 Fc region에만 결합할 수 있는 extremely high affinity 성질을 가집니다. 현재 mouse IgG1, mouse IgG2a, mouse IgG2b, rabbit IgG에 binding하는 kit들이 출시되어 있습니다. 

FlexAble labeling kits는 다음 labeling에 사용할 수 있습니다

• Primary antibodies from any supplier
• Concentrations range 0.06 mg/mL ~ 2 mg/mL
• Antibodies in any buffer formulation

FlexAble Antibody Labeling Kits는 여러 공급업체의 1차 항체를 사용하여 철저히 검증되었습니다. 저농도의 1차 항체, 50% glycerol에 저장된 항체, BSA와 같은 carriers가 포함된 항체 모두 FlexAble kits와 호환됩니다.

FlexAble Antibody Labeling Kit는 모든 항체 formulation 조건 속에서 잘 작동됩니다. High affinity FlexLinker는 그 어떤 formulation 조건에도 antibody labeling reaction 시 영향을 받지 않습니다.
Recombinant antibodies, Ascites fluid, Hybridoma supernatant formats에서도 잘 작동합니다. 

Antibody buffer formulation
FlexAble Antibody Labeling Kits는 labeling을 하기 전에 정제하거나 buffer를 교환할 필요 없이 모든 항체 formulation에서 작동합니다. 따라서 시간과 비용을 절약할 수 있고, 기존의 정제 및 buffer 교환 과정에서 발생하는 항체의 손실을 방지할 수 있습니다.
FlexAble은 항체의 Fc region에 특이적으로 결합하는 high affinity FlexLinker를 사용하기 때문에 storge buffer에 포함된 BSA, glycerol과 같은 carrier와 Tris 같은 amine계 함유 buffer 또는 sodium azide와 같은 보존제에 의한 영향을 받지 않습니다.

Anti-CD3 antibody, previously stored in the indicated concentration of glycerol, was efficiently labeled with FlexAble Antibody Labeling Kit regardless of the glycerol concentration during labeling. 1X10^6 human peripheral blood mononuclear cells (PBMCs) were stained with 0.5 µg anti-human CD3 antibody (clone UCHT1, 65151-1-Ig) labeled with FlexAble CoraLite Plus 650 Kit (KFA023) . Cells were gated for live singlet lymphocytes.

Antibody Concentration
고농도의 항체는 사전 희석 없이 사용할 수 있고, 저농도의 항체는 더 이상 농축하지 않고 사용할 수 있습니다. FlexAble은 FlexLinker의 high affinity 특징으로 인해 저농도 항체에 특히 적합합니다. (0.06 mg/mL ~ 2 mg/mL)

Standard Workflow
Labeling 0.5μg of primary antibody
Scale up proportionately to label larger quantities of primary antibody.

Before you begin, equilibrate all reagents to room temperature .

1. Combine 0.5μg of primary antibody with 1μL of FlexLinker. Add FlexBuffer to bring total volume to 8μL .
2. Mix gently by pipette for 30 sec and incubate for 5 minutes at room temperature in the dark.
3. Add 2μL of FlexQuencher.
4. Mix gently by pipette for 30 sec and incubate for 5 minutes at room temperature in the dark.
5. The antibody is now ready to be used.

Note on Multiplexing

1. 서로 다른 종의 1차 항체는 동일한 튜브에서 해당 종 별 FlexLinkers를 사용하여 동시에 labeling할 수 있습니다. Labeling할 1차 항체의 양에 따라 시약 볼륨을 적절하게 증가하여 사용합니다.
   Immunofluorescence of rat brain tissue: FFPE rat brain tissue sections were stained with anti-NeuN antibody (66836-1-Ig) labeled with FlexAble CoraLite Plus 650 Kit (KFA023, magenta), anti-TUBB3 antibody 66375-1-Ig) labeled with FlexAble CoraLite Plus 550 Kit (KFA022, orange), and CoraLite®488-conjugated GFAP antibody (CL488-60190, green).
   
   
2. 동일한 종의 1차 항체의 경우 각각 다른 vial에서 개별적으로 label을 진행합니다. 1차 항체에 lable이 완료되면 원하는 용도에 맞게 조합하여 사용하십시오.
   Immunofluorescence of HeLa: PFA-fixed HeLa cells were stained with anti-TDP43 antibody (10782-2-AP) labeled with FlexAble CoraLite® 488 Kit (KFA001, green), anti-Tom20 antibody (11802-1-AP) labeled with FlexAble CoraLite Plus 550 Kit (KFA002, red), and anti-Lamin B1 antibody (12987-1-AP) labeled with FlexAble CoraLite Plus 650 Kit (KFA003, cyan). Confocal images were acquired with a 100x oil objective and post-processed. Images were recorded at the Core Facility Bioimaging at the Biomedical Center, LMU Munich.
   
   
3. FlexAble kit로 labeling된 항체는 시중에 판매되는 conjugated primary antibody와 함께 사용될 수 있습니다.

1. FlexAble kit로 labeling 한 항체는 labeling 직후(가급적 같은 날) 사용해야 합니다.
2. Label이 부착된 항체는 약 1주 동안 보관할 수 있지만, 항체의 성능은 1일 이후 달라질 수 있습니다.
3. Labeling된 항체 보관 기간은 FlexLinker가 아닌 항체에 따라 좌우됩니다. 따라서 명확한 storage를 제안하기 어렵습니다.
