Human Cancer Models Initiative (HCMI)란 ?

HCMI는 유전자와 임상 데이터를 기반으로 차세대 Cancer Model (human tumor-derived culture model)을 개발하기 위해 설립된 국제 컨소시움으로, 현재 National Cancer Institute (NCI), Hubrecht Organoid Technology (HUB), Cancer Research UK (CRUK) Wellcome Sanger Institute (WSI) 등의 기관이 참여하고 있습니다.

연구 개발된 HCMI 모델과 데이터는 전 세계 모든 과학자들이 이용할 수 있도록 open source로 제공되며, ATCC는 개발된 HCMI 모델의 생산, 품질 검증, 보존 및 공급을 담당하고 있는 세계 유일 기관입니다.

현재 ATCC 에서는 약 250 종류 이상의 다양한 조직 유래 Organoid와 Conditionally Reprogrammed Cells (CRCs) 같은 차세대 2-D, 3-D patient-derived cancer models을 제공하고 있습니다. 희귀하거나 잘 알려지지 않은 cancer의 경우도 HCMI 컨소시움이 보유한 수많은 환자 샘플들을 통해 모델로 개발 중입니다. HCMI 모델은 기존 continuous cell line 등의 한정된 자원을 넘어 cancer에 대한 기초적 연구와 새로운 치료법의 개발 그리고 translational cancer research에 매우 유용하게 사용될 수 있습니다.

Patient-derived Next-generation Cancer Models

1. 오가노이드 모델

3-D extracellular matrix 에서 자라는 self-organizing microtissue로 pre-clinical 모델 등 다양한 연구에 활용 가능합니다.

  • May contain multiple differentiated cell types
  • Exhibit cellular polarization
  • Often possess a central lumen or other in vivo-like architecture
  • Can remain phenotypically and genotypically stable after long term expansion

2. Conditionally Reprogrammed Cells (CRCs) & Non-Organoid 모델

환자에게서 채취한 정상 또는 tumor 세포를 유전자 조작없이 빠르고 효과적인 세포 모델로 구축하는 cell culture 기술로 아래와 같은 장점을 가지고 있습니다.

  • Eliminates the need for immortalization via transduction of viral or cellular genes
  • Allows the expansion of a patient’s tumor cells
  • Reverts to differentiated phenotype in physiological culture conditions
  • Makes it possible to identify the specific mutations in these cells and to screen the cells for sensitivity to drugs
