CUT&RUN (Cleavage Under Targets & Release Using Nuclease)은 Protein-DNA 간의 상호작용을 확인하는 새롭고 강력한 기술 입니다. in vivo에서 표적 특이적 1차 항체와 Protein A-Protein G-Micrococcal Nuclease (pAG-MNase)를 이용하여 특정 protein-DNA 복합체를 분리하는 방법입니다. 기존 ChIP (chromatin-immunoprecipitation) 및 ChIP-seq에 비해 효율적으로 chromatin profiling을 가능하게 합니다. 다양한 샘플에서 histone modifications, transcription factors, transcription cofactors의 분석에 이용할 수 있으며 적은 양의 세포로 background가 적은 결과를 빠르게 얻을 수 있습니다. qPCR뿐만 아니라 NGS를 이용하여 결과를 분석할 수 있습니다.

Abstract
Cleavage Under Targets & Release Using Nuclease (CUT&RUN) is a new technology that utilizes target-specific primary antibodies and pAG-MNase to isolate protein-DNA complexes on native chromatin for analysis by qPCR or next-gen sequencing (NGS). CUT&RUN assay is a faster, more cost-effective alternative to ChIP-qPCR and ChIP-seq that requires fewer cells to investigate protein-DNA interactions. Cell Signaling Technology® (CST®) has made its CUT&RUN Kit even better by performing additional validation and optimization to improve the protocol, resulting in even more options for how you explore protein-DNA interactions.
